Abstract

This paper reviews the properties of the membrane transport of methotrexate (MTX) and the tetrahydrofolate cofactors; considers the factors that control the level of free intracellular MTX; and analyzes the possible consequences of the interaction between these substances at the level of the cell membrane that may influence the effectiveness of 'rescue' protocols. This approach may facilitate the design of more effective treatment regimens employing high doses of MTX with folinic acid 'rescue'.
