Developing a time-resolved flow cytometer for fluorescence lifetime measurements of near-infrared fluorescent proteins

Abstract

Excited state fluorescence lifetimes of near-infrared fluorescent proteins iRFP682 and iRFP713 engineered from bacterial phytochromes were measured with a time-resolved flow cytometer. Average lifetimes ranged between 0.5 and 1.0 ns for n=5000 bacterial cells. We anticipate cell sorting for screening cells expressing mutated iRFP variants of high quantum yield.
