This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The study will address the potential impact of diazoxide, which acts via a central mechanism, on inhibiting endogenous glucose production in the liver, and to investigate its use as treatment for diabetes melitus. We will determine the effect of diazoxide on hepatic glucose production in nondiabetic, healthy, young individuals under fixed hormonal conditions. Rates of glucose production will be measured during "pancreatic clamp" studies, with somatostatin infusion and basal hormone replacement in the presence of normoglycemia (plasma glucose ~90 mg/dl) under experimental conditions.
